Samuel L. Clemens first used the name "Mark Twain" in February 1863 while working for the Virginia City Territorial Enterprise. 

There was a "Pony Express" in Virginia City but it was unrelated to the transcontinental Pony Express. See http://www.xphomestation.com/vcxp.html for more information on the Wells Fargo version.
